Hampshire elected to bat, and appeared to regret that decision as they slumped to 21 for three against Devon's opening attack of Kaz Szymanski and Matt Parker.
The decision was vindicated with a 157-run partnership between Alex Willoughby (73) and Patrick Holly (75).
From a strong position of 178 for three, Parker (right) was re-introduced with 15 overs remaining, and produced a stunning spell of six for 17 in six overs reducing the visitors to 214 for nine.
A last wicket flourish Tom Spencer (14) and number 11 Scott Stuart (16) added 31 runs to take the score to 245 all out in the final over.
Parker ended up with career-best Devon figures of seven for 38 from his 10 overs - a feat made more remarkable following his recent return from a back stress fracture injury that ruled him out of the South Africa tour in February.
Parker was well supported by Szymanski with economical figures of two for 19 off 10.
Devon were banking on a solid start from openers and Exeter colleagues Jamie Khan and James Bovey and were not disappointed.
The openers posted 81 runs in 15 overs, with Khan in particular good nick with eight fours in his 45 before he chipped back a caught and bowled.
Bovey played the supporting role and reached 34 with three fours before his skipper Tom Lammonby ran him out.
Lammonby continued in his normal, stylish way and added 49 for the third wicket with Sam Maunder, before Maunder was caught on 25.
Lammonby hit fours fours in his score of 57 before getting out.
Devon were always slightly ahead of the run rate, helped by a brief 15 from Mikey Cooper.
Szymanski guided Devon home with a controlled 30 not out, supported by Jack Gibbs (5no).
